Although the molecular links underlying the causative relationship between chronic low-grade inflammation and chronic diseases development are not completely understood, compelling evidence suggests a pivotal role for specific inflammatory cascades. The main aim of the present activity is to provide to PhD students recent emerging evidence on the experimental and clinical approaches to counteract the the development of a chronic inflammatory status thus contributing to limit the pathogenesis of many relevant progressive and degenerative diseases.

"Pharmacometrics: Modeling & Simulation in Pharmacology - 1"

Pharmacometrics has become a term that encompasses modeling & simulation for pharmacokinetics, pharmacodynamics, exposure–response relationship, and disease progression. Pharmacometrics lies at the heart of what drug companies do: collecting data from animals, normal volunteers, and patients; quantifying it, and then being able to determine what that data mean for optimizing drug efficacy and minimizing toxicity. Pharmaceutical and biotech companies have invested heavily in establishing pharmacometrics expertise, to utilize the preclinical, clinical data to understand the disease progression, the drug behavior, and its effect on individual patients and to personalize medicine to specific groups of patient population. The purposes of this lesson is to provide an overview of different approaches that were used in pharmacometrics in the context of pharmaceutical drug development.

The promising future of light and ultrasound in pharmacology

Light and ultrasound (US) are fascinating phenomena that tie together physics, chemistry, and biology.
Various pharmacological strategies relying on the use of light have emerged during the last decades. In particular, photopharmacology is a novel approach that relies on the use of reversibly photoswitching target-specific drugs that change their efficacy upon irradiation and allow for the control of defined biological targets. Recent development of therapeutic US applications refers to the use of US energy for increasing drug efficacy. US energy can be tuned to create changes in tissue permeability for enhancing the delivery of conventional drugs, as well as for enhancing the drug delivery via specially tailored vehicles or for activating responsive drugs.

Breakthrough in cancer immunotherapy

Cancer immunotherapy essentially refers to the attempt of utilising and boosting natural immune responses, to combine external therapy (chemo-/radiotherapy/surgery) with the internal defence and control machinery at hand, thereby maximizing treatment efficiency. Researchers shifted the paradigm from attempting to activate the immune system (i.e., vaccinating) to releasing the checkpoints that keep it in a negative regulatory mode; checkpoint-blockade immunotherapy has arguably been the most exciting advance made in cancer treatment in recent years.

"Pharmacometrics: Modeling & Simulation in Pharmacology - 2"

A key challenge in early clinical investigation of novel drugs is their inherent risk of adverse effects. Although some adverse effects may be acceptable based on the risk/benefit ratio to the patients, these should be avoided at the starting dose of a first-in-human (FIH) study, particularly when normal healthy volunteers are the study subjects. As a result, determining a safe starting dose is an important first step in preparing new agents for FIH studies. In the past decade, the methods of the FIH starting dose selection have evolved from mostly toxicology-based approaches to a more integrated pharmacology- and mechanism-based method, such as determining a minimal anticipated biological effect level (MABEL) as the FIH starting dose. This lesson will intend to provide an overview of the evolution of FIH starting dose selection approaches and to review the principles and procedures of more integrated pharmacology- and mechanism-based methods for the MABEL determination.
